The prophecy of Esau coming to kill Jacob foretells the later conflict in Revelation, where Jewish Israel is surrounded by enemies, and as they are raped and killed, they call out for Jesus as the Messiah, who will come to save them.
Based on my research of the Bible and related commentaries, the prophecy about the conflict between Esau and Jacob does foretell a long-term, spiritual struggle, foreshadowing what is to come as well.
